A high fidelity restriction endonuclease that recognizes the sequence G^GTNACC. High Fidelity (HF) Restriction Enzymes have 100% activity in rCutSmart Buffer; single-buffer simplicity means more straightforward and streamlined sample processing. HF enzymes also exhibit dramatically reduced star activity. HF enzymes are all Time-Saver qualified and can therefore cut substrate DNA in 5-15 with the flexibility to digest overnight without degradation to DNA. Engineered with performance in mind, HF restriction enzymes are fully active under a broader range of conditions, minimizing off-target products, while offering flexibility in experimental design. NEB extensively performs quality controls on all standard and high-fidelity (HF) restriction enzymes. Examples of nuclease contamination studies for some of our HF restriction enzymes are shown below.
EPICYPHER INC CUTANA pAG-Tn5 for ChIC/CUT&Tag, 50 rxns
Recombinantly produced in E. coli, CUTANA pAG-Tn5 for ChIC/CUT&Tag is a fusion of Proteins A and G to Transposase Tn5. This construct is useful in performing Cleavage Under Targets and Tagmentation (CUT&Tag). The active dimer of Transposase Tn5 is charged with Illumina adapters and ready to be used immediately in CUT&Tag. CUTANA pAG-Tn5 does not contain an epitope tag, which makes it compatible with tag-mediated CUT&Tag.

EnGen Spy Cas9 NLS, is an RNA-guided endonuclease that catalyzes site- specific cleavage of double stranded DNA. The location of the break is within the target sequence 3 bases from the NGG PAM (Protospacer Adjacent Motif) ). The PAM sequence, NGG, must follow the targeted region on the opposite strand of the DNA with respect to the region complementary sgRNA sequence. EnGen Spy Cas9 NLS contains Simian virus 40 (SV40) T antigen nuclear localization sequence (NLS) on the N- and C- termini of the protein.

New England Biolabs, Inc. Quick CIP – 5000 units
Quick CIP is a heat-labile recombinant version of calf intestinal alkaline phosphatase (CIP) purified from a recombinant source. Quick CIP nonspecifically catalyzes the dephosphorylation of 5 and 3 ends of DNA and RNA phosphomonoesters. It also hydrolyses ribo- and deoxyribonucleoside triphosphates (NTPs and dNTPs). Quick CIP is useful in many applications that require the dephosphorylation of DNA or RNA ends. In cloning, dephosphorylation prevents re-ligation of linearized plasmid DNA. The enzyme can quickly dephosphorylate 5 protruding, 5 recessed, and blunt ends in just 10 minutes. Quick CIP may also be used to degrade unincorporated dNTPs in PCR reactions to prepare templates for DNA sequencing or SNP analysis.
